Diethyl ethoxymethylenemalonate is applied in the synthesis of pyrido[3,2-e]pyrimido[1,2-c]pyrimidines. It was first used to detect lysine decarboxylase activity. It supports amino acid analysis via precolumn derivatization in reversed‑phase HPLC. It is a key reagent in the Gould Jacobs reaction for quinoline synthesis, such as forming 4-hydroxyquinoline from aniline. It also serves as an intermediate for manufacturing flumequine, a fluoroquinolone antibiotic.
